The regeneration of these plants is carried out by in vitro induction of embryogenesis in microspores and pollen grains. This process is switched by the application of stress treatments and hormones, but the efficiency is still very low in many crops. The molecular and cellular processes responsible of the change in the developmental program of the microspore are still under investigation. Defined ultrastructural and expression changes have been reported to accompany the reprogramming of the microspore to embryogenesis in dicot systems (Testillano et al., 2000), but less is known on the cellular characterization of the process in monocots.

P-88: Assessing Expression Changes of Some Wnt Pathway Genes During Goat Early Embryonic Development
Background: The developmental competency of embryos is affected by several factors, including the developmental pathways and their elements. In mammalian species including goat, fertilized oocyte undergoes several divisions to form a structure called blastocyst. These events depend on the successful control of temporal and spatial expression of genes involved in genome activation. One of the cr...
متن کاملExpression profiles in barley microspore embryogenesis
In barley, microarray technology has allowed to study the gene expression profiling associated with the stress pretreatment phase of microspore embryogenesis, where the reprogramming of microspores takes place. Transcriptome analysis of anthers before and after four days of mannitol pretreatment, revealed changes in the expression of 2,673 genes. A thorough study of these differentially express...
متن کاملTranscriptome analysis of barley anthers: effect of mannitol treatment in microspore embryogenesis
Carbohydrate starvation is an efficient stress treatment for induction of microspore embryogenesis. Transcriptome analysis of anthers response to mannitol treatment using the 22k Barley1 GeneChip revealed large changes in gene expression. Statistical analysis and filtering for 4-fold or greater changes resulted in 2,673 genes, of which 887 were up-regulated and 1,786 down-regulated. Great diffe...

Induction of embryogenesis from isolated microspore cultures is a complex experimental system where microspores undergo dramatic changes in developmental fate. After ~40 years of application of electron microscopy to the study of the ultrastructural changes undergone by the induced microspore, there is still room for new discoveries. In this work, high pressure freezing and freeze substitution ...
متن کاملChanges in DNA methylation levels and nuclear distribution patterns after microspore reprogramming to embryogenesis in barley.
Under specific stress treatments, the microspore can be induced in vitro to deviate from its gametophytic development and to reprogram towards embryogenesis, becoming a totipotent cell and forming haploid embryos. These can further regenerate homozygous plants for production of new isogenic lines, an important biotechnological tool for crop breeding. DNA methylation constitutes a prominent epig...
